Methods of asepsis for in vitro establishment and germination of Eucalyptus grandis

Many technical and scientific studies have been carried out for eucalyptus cultivation, especially in the area of large-scale clonal propagation. However, in some studies it is necessary to obtain aseptic materials via seeds, and for this purpose there are no efficient aseptic protocols. Given this problem, the present study aimed to define a protocol for asepsis of Eucalyptus grandis seeds for establishment and germination in vitro. Four aseptic methods were tested using benomyl-based fungicide, hydroalcoholic solution (70% v / v) and sodium hypochlorite (NaOCl), alternating the order of application. The experiment was conducted in a completely randomized design with five replications. The percentage of establishment and contamination were evaluated at the end of the experiment. The best method found was the sterile water wash sequence followed by 70% alcohol for 30 s, 2.5% sodium hypochlorite for 20 min, 1% benomyl fungicide for 20 min and three washes with sterile water, corresponding to 87.5% of establishment.
