The Citrus Canker (CC) caused by Xanthomonas citri subsp. citri (Xcc), is responsible for severe damage to cultivated Citrus plants worldwide. The selection of resistant genotypes is the most effective and environmentally sustainable way to manage CC, providing long-term solutions. Breeding for disease resistance and, consequently, genotype selection requires efficient screening techniques. This study aims to identify an inoculation method that is easy to use on a large scale and responsive to screening for CC-resistant genotypes under greenhouse conditions. The study was conducted under a greenhouse at the Núcleo de Pesquisa em Biotecnologia Aplicada in two replicates from January to March of 2017 and 2018. Three inoculation methods were performed with five bacteria concentrations. The activities of three antioxidant enzymes were evaluated in the second experiment. The results indicated that needle wound inoculations with concentrations between 1.39x105 and 1.52x105 CFU ml-1 to inoculate Xcc were the most favorable to disease progress. Catalase e Peroxidase showed a significant increase of activity over time among treatments, especially to needle wound with bacteria.
