Abstract

Micropropagation is an important tool for the genetic improvement of several tree species and, for the genus Citrus, it offers great potential for the in vitro production of high-quality rootstocks. The success of micropropagation is influenced, among other factors, by the type and concentration of growth regulators added to the culture medium. Thus, the objective of this study was to evaluate the effects of different cytokinins on the induction and growth of shoots, as well as the effects of different concentrations of IBA on the induction and formation of adventitious roots in nodal segments of Rangpur lime (Citrus limonia Osbeck), excised from seedlings germinated in vitro. For shoot induction, the cytokinins BAP, KIN and TDZ were tested, added to the WPM culture medium at a concentration of 1,0 mg.L-1, and for the induction and formation of adventitious roots, the concentrations 0,5; 1,0; 1,5 and 2,0 mg.L-1 of the auxin IBA added to the WPM medium. The superiority of BAP as a sprouting inducer was observed, and its addition to the culture medium provided a higher percentage of sprouted explants (87.5%) and a higher average number of sprouts per explant (1.75). The TDZ induced hyperhydration in 91.7% of the explants. The concentration of 1.5 mg.L-1 of IBA in the nutrient medium is the most indicated for the formation of adventitious roots, promoting a higher percentage of rooting (100%), average number of roots per explant (2.83) and average root length (42.74 mm).
